Selection of a fast-growing Chikungunya virus by passage from suckling mouse brain to VERO cells

Resumo: Chikungunya seed viruses passaged through suckling mouse brains propagated very slowly in VERO cells at the first passage, whereas the fourthly VERO cell-passaged virus grew rapidly in the cultured cells. This was evidenced by the quantitative assay of plaque-forming unit and the mode of viral RNA synthesis. The slow growth of the virus at the first passage was due to neither insufficient adsorption of the virus to VERO cells, nor influence of mouse brain substances included in the virus seed, nor that of a product of the infected cells, e. g. interferon.
