Characterization and expression of a novel holotype insecticidal crystal protein gene from native Bacillus thuringiensis BM59-2

Resumo: We characterized a novel holotype cry gene (cry52Ba1) harbored in a Bacillus thuringiensis isolate BM59-2 native to China that expresses a spherical crystal protein of about 80 kDa. In this study, the full length of the cry52Ba1 gene was cloned from this strain. Sequence analysis of the gene was also performed. Furthermore, cry52Ba1 was expressed in Escherichia coli BL21(DE)pLysS, and the resulting insecticidal activity showed that the Cry52Ba1 protein exhibited high larvicidal activity against Aedes aegypti (Diptera), with a lethal concentration 50 of 1.526 kg/mL (95% confidence: 0.740-3.499 kg/mL).Original Abstract: Nous avons caracterise un nouveau gene holotype cry (cry52Ba1) contenu dans l'isolat BM59-2 de Bacillus thuringiensis, originaire de Chine, qui exprime une proteine d'environ 80 kDa. Dans cette etude, le gene cry52Ba1 de pleine longueur a ete clone a partir de cette souche et la sequence du gene a ete determinee et analysee. De plus, cry52Ba1 a ete exprime chez Escherichia coli BL21(DE)pLysS et le resultat de la mesure de l'activite insecticide a montre que la proteine Cry52Ba1 possedait une forte activite larvicide envers Aedes aegypti (Diptere), avec une concentration letale a 50 % (CL sub(50)) de 1.526 kg/mL (95 % de confiance: 0.740-3.499 kg/mL).
